Excipients are pharmacologically inactive substances that are generally used as a carrier of the API or active ingredient in the drug. Pharmaceutical excipients originate from a variety of sources and include animal origin (lactose, gelatine, stearic acid, beeswax, honey musk, lanoline), plant origin (starches, sugars, cellulose, alginates), mineral origin (talc, calcium phosphate, silicon dioxide) and by synthesis (povidone, polysorbates, polyethylene glycol). Although excipients do not have any therapeutic effect themselves, they play crucial roles in the manufacturing process and the performance of the final drug product. Excipients can help enhance drug stability, improve solubility, aid in drug absorption and distribution, provide taste/mask unpleasant flavours, and ensure the proper release of the drug. Here are five key product groups (amongst others) of pharmaceutical excipients commonly used today, and regularly sold by CARBOCRAFT:

Binders
  • These excipients help bind together the powder particles of the drug formulation, ensuring the tablet or capsule retains its shape. Common binders include cellulose derivatives (e.g., hydroxypropyl cellulose), polyvinylpyrrolidone (PVP), and starch.
Coatings
  • Coatings are used to provide a protective layer around tablets or capsules, which can facilitate swallowing, mask unpleasant odours or tastes, and control the release of drugs. Common coating agents include hydroxypropyl methylcellulose (HPMC), ethyl cellulose, and sugar-based coatings like sucrose or glucose.
Disintegrants
  • These excipients aid in the disintegration or breakup of a tablet or capsule when it comes into contact with body fluids. By promoting rapid disintegration, disintegrants facilitate drug dissolution and absorption. Common disintegrants include croscarmellose sodium, crospovidone, and sodium starch glycolate.
Fillers and diluents
  • These excipients are used to increase the bulk volume of a tablet or capsule. Fillers and diluents help achieve consistent dosing and facilitate tablet compression. Examples include lactose, microcrystalline cellulose, and calcium phosphate.
Lubricants and glidants
  • Lubricants reduce friction between the tablet/capsule formulation and the processing equipment during manufacturing. Glidants improve the flow properties of powders, ensuring uniform filling of capsules. Examples of lubricants and glidants include magnesium stearate, stearic acid, and silica.

Pharmaceutical APIs (Active Pharmaceutical Ingredients), also known as drug substances, are the biologically active components responsible for the therapeutic effects of a drug. They are the main active compounds that provide the desired pharmacological action in the body. APIs are typically derived from natural sources or synthesized through chemical processes. Here are some key examples:

  • Enzymes: Enzyme APIs are biocatalysts that facilitate biochemical reactions in the body. They are used in various therapeutic settings, diagnostics, and industrial processes. Enzyme APIs may include thrombolytic enzymes used to treat blood clots, digestive enzymes for enzyme replacement therapy, and enzyme-based diagnostic reagents for detecting specific biomarkers.
  • Hormones: Hormonal APIs are substances that mimic or regulate the functions of natural hormones in the body. They are used for hormonal replacement therapy, contraception, fertility treatment, and various endocrine disorders. Hormonal APIs include insulin, estrogen, progesterone, testosterone, and thyroid hormones.
  • Macromolecules and Biologics: These APIs are larger molecules, often derived from living organisms and manufactured using biotechnological processes. They include proteins, peptides, antibodies, and nucleic acids. Macromolecule APIs may have complex structures and are used to treat various diseases, such as insulin for diabetes, monoclonal antibodies for cancer treatment, and interferons for antiviral therapy.
  • Small Molecules: This group comprises most traditional drugs in the pharmaceutical market. Small molecule APIs have a low molecular weight and are chemically synthesized. They often target specific receptors or enzymes in the body to exert their therapeutic effect. Examples include aspirin (acetylsalicylic acid), paracetamol (acetaminophen), ibuprofen, and statins used in cholesterol management.
  • Vaccines: Vaccines are preparations containing weakened or killed microorganisms, antigens, or recombinant proteins that help stimulate the immune system and provide immunity against infectious diseases. Examples of vaccine APIs include those used for influenza, hepatitis, measles, mumps, rubella, and more. The composition of vaccine APIs can vary widely depending on the specific disease they target.
